Deadpool was more about the jokes and genre defying moments than plot, therefore I believe the plot was actually very short and almost insignificant as the allure of the movie was how much it poked fun at the longstanding super hero genre.

Therefore, for the most part, the first act is back story (funny and enjoyable but back story none the less) only after he is given the option to cure the cancer does his story begin. This is the motivating event that pushed him to pursue and destroy the bad guy.

After a terminally ill gun for hire is cured and given healing super powers but tortured and disfigured in the process, he manages to break free and hunt down his tormentor.
